What is Conolidine

Conolidine is widely considered one of the greatest pain relief discoveries in the last 20 years. It is one of the 66 alkaloids found in the Tabernaemontana divaricata (Crepe Jasmine) plant; generally from the bark and root of the plant.
Conolidine has been tested extensively as a pain reliever and has been compared in efficacy to more traditional, opiate-derived pain medications like morphine. Early scientific opinion surmises that Conolidine is the first known alkaloid to clear the “Scavenger” receptors in your brain, potentially allowing for your body’s natural endorphins to be fully optimized and regulated; a true first of its kind.

How It Works

CONOCB2™ works on multiple functions of the brain and body. Tabernaemontana divaricata (Crepe Jasmine), which contains the Conolidine alkaloid, works on a fairly recently discovered opioid receptor named ACKR3/CXCR7 and regulates naturally produced opioid peptides in the brain.2

What does that mean? This means that the body’s naturally produced painkillers (opioid peptides) are more capable of reaching your brain’s opioid receptors instead of being “scavenged.” Conolidine essentially creates a barrier between the “Scavenger” Receptors and Opioid Receptors, allowing your body’s ability to deliver its own endorphins to be fully optimized. In addition to the Crepe Jasmine, the Palmitoylethanolamide (PEA) targets alpha peroxisome proliferator-activated receptors (PPAR-α) found on neurons in the brain that are naturally activated as a result of nerve damage, along with targeting cannabinoid receptors (CB1, CB2).3, 4
Conolidine also affects transient receptor potential vanilloid type-1 (TRPV1) ion channels which have been shown to compound the effects of the Crepe Jasmine and increase anti-inflammatory effects. Finally, the Piperine (Black Pepper Extract 95%) dramatically increases bioavailability, a common issue with Dietary Supplements and other medications. Most predominantly, Piperine has been shown to increase absorption in the GIT (gastro-intestinal tract) by inhibiting or inducing numerous enzymes including, but not limited to, CYP1A1, CYP1B1, CYP1B2, CYP2E1, CYP3A4 etc.5

When administered orally, CONOCB2™ will work into the body via the mucous membranes in the GI Tract, mouth and under the tongue (sublingual), travel to the brain and is able to pass through the blood-brain barrier due to the use of our proprietary extraction method used when producing CONOCB2™.

 

This method of delivery allows for relatively fast absorption of compounds that may not be generally as effective by standard GI ingestion.

Transdermal

When administered to the skin, CONOCB2™ has the ability to travel into the bloodstream as a result of the same solvents that make it so effective when Orally administered.

The Entourage Effect is a fairly new concept that simply suggests that the sum of the whole is far more effective than its parts. For example, it could be argued that a standard hydrocodone pain pill containing 500mg of acetaminophen and 10mg of hydrocodone is far more effective than either the 500mg of acetaminophen or the 10mg of hydrocodone alone. In the case of CONOCB2™, the same approach is taken.
